1749年4月30日设县。县名来自南安普顿区：在殖民地时期区在郡之下，由一百家或可以提供一百个有战斗力男性的家庭组成。南安普顿指的是亨利·罗赛斯雷，第三代南安普顿伯爵 (Henry Wriothesley,the 3rd Earl of Southampton)。
