颅骨锁骨发育不良症是控制成骨细胞特殊转录因子发生基因突变的疾病，导致头骨骨化慢，骨缝密合极慢，可能到4岁才开始骨化，甚至到成年还未完成。典型时会造成头大、脸小、肩下垂及胸部狭窄。

其发生率为1/1000000。

遗传方面，其遗传方式为体染色体显性遗传模式，无关性别，约2/3患者有家族遗传。
