痘病毒(Poxviridae，pox=发疹的病)为哺乳动物的最大及最复杂的病毒(170×950nm)。包含两个亚科痘病毒脊索亚科 (Chordopoxvirinae)、痘病毒昆虫亚科 (Entomopoxvirinae)，这两个亚科各有8个和4个属。不同的动物有不同品系的病原病毒。遗传物质为双股DNA，长度约130-375knt; 病毒直径约140-260 nm，最大的病毒就是属于痘病毒科。通常可区分为8属(genera)，分别为正痘病毒属 (Orthopoxvirus)、骆驼痘病毒属 (Capripoxvirus)、猪痘病毒属 (suipoxvirus)、兔痘病毒属 (Leporipoxvirus)、软件动物痘病毒属 (Molluscipoxvirus) 、雅塔痘病毒属(Yatapoxvirus)、禽痘病毒属 (Avipoxvirus)、副痘病毒属 (Parapoxvirus)，其中两属(Orthopoxvirus及Parapoxvirus)为人类的重要病原。同一属病毒的血清反应常有广泛的交叉反应，但各属间仅有一种共同抗原，此抗原一般认为是一种核蛋白复合体。天花的病原Variola病毒为Orthopoxvirus属，此病毒曾经摧残全人类，目前已被扑灭，然最近常被认为为一种有可能之再新兴之疾病。其他的痘病毒如骆驼痘(camel pox)及ectromelia造成低等动物的皮肤发疹。天花(Variola)及传染性软疣(molluscum contagiosum)病毒只对人类有病原性。
